Description

E2F transcription factors are important key regulators of cell growth control. There are 5 types of E2F transcription factors (E2F1-E2F5) that play an important role in a number of pathological conditions (1). A double mutant of E2F1/E2F2 mouse has shown to develop non-autoimmune insulin-deficient diabetes and exocrine pancreatic dysfunction with exocrine dysplasia (1). The E2F family of transcription factors consists of two subgroups termed E2F and DP transcription factors that binds DNA cooperatively with dp proteins through the E2 recognition site, TTTCC/GCGC, found in the promoter region of a number of genes. E2F1 is a member of the E2F transcription factors (cell cycle Genes) that are key components of the cyclin D/retinoblastoma/E2F pathway (1). E2F1 binds DPARBR1 proteins. The eukaryotic E2fs are conserved constriction factors which serve an antagonistic role in several pathways related to cellular division, DNA repair, and differentiation (2). Therefore, E2F1 is expressed throughout the cell cycle. E2F1 is one of the key targets for axon to determine whether cells proliferate or whether they exit the cell cycle, enlarge and endoreduplicate their DNA (2). E2F proteins are also involved in the regulation of human intestinal epithelial cell proliferation and development (3). The expression of E2F1 is decreased and E2F4 protein is sequestered in the cytoplasm during G(0) arrest associated with serum deprivation, confluence, and terminal differentiation of intestinal cells (3). E2F1 deficiency also impairs murine spermatogenesis and augments testicular degeneration. Abundance of E2F1 is increased by auxin trough stabilization of the protein (4). CDK levels are increased by E2F1 even in hormone free conditions. E2F1 activity was shown to induce S phase and endoredupolication. Over expression of E2F1 in transgenic plants lead to modifications of the morphology of the roots, cotyledons, and leaves that can be ascribed to stimulation of cell division. These plants had increased levels of E2F1 as well as ATE2Fb protein which suggests that E2F1expression might be upregulated by Ate2Fa transcription factor. E2F1 is approximately a 56kDa protein, 437 amino acids. The E2F1-selective antibodies were generated against three antigenic sequences form N-, midregion and C-terminal end of the E2F1 protein using peptide corresponding to human E2F1 protein. The antibodies E2F1 are affinity purified over immobilized antigen based chromatography, and the purified immunoglobulins are stabilized in antibody stabilization buffer.
